Advanced Method Development for HPLC/UHPLC (2 Days)

25 May 2017 08:45-17:00, Manchester, United Kingdom

Introduction
Who should take this course?
If you’ve ever wished that HPLC or UHPLC method development was more logical and methodical,
this is the course for you. This course is the ‘gold standard’ in LC method development courses
and is for anyone who has some involvement in developing new methods, troubleshooting older
methods or transferring existing methods into a routine laboratory. Attendees should have at least
some LC experience in the laboratory.
What does it cover?
This two day course focuses on the fundamentals of chromatographic separation and applies them
in a time-proven strategy, using the principles of QbD to enable faster development of better, more
robust HPLC and UHPLC methods. This two day course includes a number of interactive workshops
and discussions that encourage a thorough understanding of the topics covered. Delegates will be
introduced to LC method development software and shown how it can be used to help them find
the optimum separation conditions. During the course, delegates will solve a series of real-world
LC method development problems using chromatographic computer modelling, demonstrating
how effectively this approach to method development works. The principles used in the computer
modelling examples are fully applicable and translatable to the real-world separations and
separation strategies in your laboratory.
What will I get from this course?
Based on the instructor’s extensive experience in the laboratory supporting pharmaceutical and
bioanalytical method development, those attending this course will gain the practical skills needed
to develop better HPLC and UHPLC methods in a shorter time. The important principles of
resolution will be explored in order to provide a solid platform from which delegates will be able
to implement a systematic, efficient approach to developing rugged, robust reversed-phase LC
methods. The techniques can be used as a stand-alone method development strategy or added
to existing procedures to help streamline method development processes. On this course you will
be shown how to achieve the best possible chromatography, save hours of frustration and improve
your results time after time.
Quality by Design?
The central tenet of Quality by Design (ICH Q8) is that quality cannot be tested into a product –
instead it must be designed into the product. When the product is an HPLC or UHPLC method,
QbD strategies can be used to guide the method development process resulting in a standardised
procedure, that provides better methods more quickly, that are more easily validated and are easier
to use and adjust in routine applications afterwards.
Course Outline – Day 1
Introduction to LC method development
HPLC/UHPLC basics
HPLC/UHPLC columns
Reversed-phase separations for neutral molecules
Reversed-phase separations for ionic samples
Equivalent and orthogonal columns
Course Outline – Day 2
Gradient elution
UHPLC Considerations
Quality by Design
Normal-phase and HILIC
Day 2 Bonus Options for the course:-
Chiral, Ion-exchange and prep
Quality issues
LC-MS & LC-MS/MS
Sample preparation
During the course the core sections will always be covered. Then, depending on the interests of
the particular group of attendees and the remaining time, one or more of the Bonus topics may be
included.
The main goal of this course is to provide valuable, practical help to attendees that can be applied
in real situations in real laboratories. Delegates are encouraged to bring questions, separation
problems and other topics of interest to discuss during the course as a group or one-to-one with
the instructor.
